Primary Abstract & Repository Description

Source: Zenodo (CERN Open Data)
Version numbering — the public record jumps from version 4 (2026-09-30) to this release. The deposit was revised twice after v4 without being published (internal v5, 2026-10-19; internal v5.2, 2026-10-20); this release (internal v5.3, 2026-10-26) subsumes both and is what Zenodo displays as version 5. Versions 5 and 5.2 below are therefore internal drafting history, not public releases. Version 5.3 (2026-10-26) — the self-written MR-PRESSO analog was replaced by the official implementation (rondolab/MR-PRESSO, 5,000 distributions, seed 42; code/c29_mpresso_official.R, results/tables/c29_mpresso_official.csv). Global-test p-values: 0.060 (ischemic stroke), 0.071 (any stroke), <2e-4 (AD, Bellenguez), 0.001 (MDD), 0.216 (AD, Kunkle); both stroke outcomes become non-significant with zero outliers, so stroke headline estimates are the raw IVW values. The official corrected AD estimate is OR 0.813 (p = 0.0024); raw and corrected estimates are now reported side by side. The deposited manuscript is submission version 2 (2026-10-26), and Tables S3/S4 plus the abbreviations were aligned to the official MR-PRESSO values. Version 5.2 (2026-10-20; internal draft, never published)...

Editorial Critique, Methodological Value & Reusability Guide

Structured via CNS Domain Knowledge Engine

1. Scientific Context & Premise

Primary Premise (Zenodo (CERN Open Data)): "Version numbering — the public record jumps from version 4 (2026-09-30) to this release." Within Alzheimer's Disease (AD), this work examines Zenodo, Open Dataset, Mendelian randomization, coronary artery disease, stroke in relation to Aβ42/40, p-tau217, p-tau181, GFAP, NfL, and APOE ε4 genotype interactions. Authored by Fang, Xiaoling, Zhang, Xue, Shi, Shuguang.

2. Study Architecture & Findings

Study Architecture: Experimental / Clinical CNS Design (1 Data Files) utilizing RNA-seq [DOI: 10.5281/zenodo.23653594]. Reported quantitative parameters in the abstract include: p = 0.0024. Key reported conclusion: "Version 5.2 (2026-10-20; internal draft, never published)..."

3. Secondary Analysis & Reusability

Secondary Analysis Applications: (1) Cross-referencing RNA-seq variables (Zenodo, Open Dataset, Mendelian randomization, coronary artery disease, stroke) against open repositories in Alzheimer's Disease (AD); (2) Benchmarking feature distributions and effect sizes reported in "Colocalization-filtered Mendelian randomization of coronary artery disease and s"; (3) Informing cohort stratification and endpoint selection.

4. Methodological Caveats & Governance

Methodological Note: When integrating these RNA-seq findings, account for cohort-specific inclusion criteria (1 Data Files), potential site/instrument batch effects, and repository license terms (MIT-LICENSE).

query = "DOI:\"10.5281/zenodo.23653594\""
url = f"https://www.ebi.ac.uk/europepmc/webservices/rest/search?query={urllib.parse.quote(query)}&format=json&resultType=core"

with urllib.request.urlopen(url, timeout=10) as response:
    payload = json.loads(response.read().decode("utf-8"))
    results = payload.get("resultList", {}).get("result", [])
    if results:
        record = results[0]
        print("Title:", record.get("title"))
        print("Journal / Source:", record.get("journalTitle") or record.get("bookOrReportDetails"))
        print("Cited By Count:", record.get("citedByCount", 0))

Standardized 4-Step Workflow & Governance Checklist

Standardized Acquisition & Preprocessing Workflow

1. Programmatic Metadata & Full-Text OA Retrieval

Query Europe PMC or Crossref REST APIs using the persistent DOI/URL to retrieve structured XML full text, supplementary tables, and grant links.

2. Study Design & Cohort Eligibility Screening

Extract sample size, inclusion/exclusion criteria, and primary outcome endpoints for systematic review (PRISMA) inclusion.

3. Data Availability & Supplementary Repository Linkage

Locate accession numbers (GEO, SRA, OpenNeuro, DANDI, Zenodo, Figshare) referenced in the Data Availability section.

4. Citation Graph & Replication Tracking

Monitor supporting and contrasting citations over time to verify whether key findings have been independently replicated.

Reproducibility & Governance Checklist

  • Verify whether the source is a peer-reviewed journal publication or an unrefereed preprint before clinical citation.
  • Check supplementary data licenses (CC-BY / CC0) when extracting figures or summary statistics for meta-analyses.
  • Cross-reference NCT clinical trial identifiers when evaluating therapeutic efficacy or biomarker endpoints.
