Clinical Dementia Rating — Sum of Boxes
臨床認知症評価尺度 ボックス合計点(CDR-SB)
1. Biological & Technical Mechanism
The Clinical Dementia Rating Sum of Boxes (CDR-SB) is calculated by scoring six cognitive and functional domains—Memory, Orientation, Judgment & Problem Solving, Community Affairs, Home & Hobbies, and Personal Care—each on a 5-point scale (0, 0.5, 1, 2, 3) based on semi-structured interviews with both the patient and a reliable study partner. Summing the six domain scores yields a continuous 0–18 scale that is highly sensitive to subtle progression from Mild Cognitive Impairment (MCI) to mild Alzheimer's dementia.
2. Open Science & Clinical Trial Significance
CDR-SB served as the primary efficacy endpoint in pivotal Phase 3 trials of anti-amyloid antibodies (such as Lecanemab / Clarity AD) and is extensively documented in ADNI and Alzforum therapeutics trackers.

Agora Target Validation: INPP5D (inositol polyphosphate-5-phosphatase D) in Alzheimer’s Disease
This gene is a member of the inositol polyphosphate-5-phosphatase (INPP5) family and encodes a protein with an N-terminal SH2 domain, an inositol phosphatase domain, and two C-terminal protein interaction domains. Expression of this protein is restricted to hematopoietic cells where its movement from the cytosol to the...
Agora Target Validation: NRN1 (neuritin 1) in Alzheimer’s Disease
This gene encodes a member of the neuritin family, and is expressed in postmitotic-differentiating neurons of the developmental nervous system and neuronal structures associated with plasticity in the adult. The expression of this gene can be induced by neural activity and neurotrophins. The encoded protein contains a ...
Agora Target Validation: BIN1 (bridging integrator 1) in Alzheimer’s Disease
This gene encodes several isoforms of a nucleocytoplasmic adaptor protein, one of which was initially identified as a MYC-interacting protein with features of a tumor suppressor. Isoforms that are expressed in the central nervous system may be involved in synaptic vesicle endocytosis and may interact with dynamin, syna...
Allen Brain Atlas: Multi-plane optical physiology during image change detection
Time series of behavioral variables and neural activity measured with multi-plane in vivo 2-photon imaging in mice performing a visual change detection task where mice were trained with image set A and tested with a novel image set B... Freely accessible open data product maintained by the Allen Institute for Brain Science.
Agora Target Validation: HTRA1 (HtrA serine peptidase 1) in Alzheimer’s Disease
This gene encodes a member of the trypsin family of serine proteases. This protein is a secreted enzyme that is proposed to regulate the availability of insulin-like growth factors (IGFs) by cleaving IGF-binding proteins. It has also been suggested to be a regulator of cell growth. Variations in the promoter region of ...
Agora Target Validation: APOE (apolipoprotein E) in Alzheimer’s Disease
The protein encoded by this gene is a major apoprotein of the chylomicron. It binds to a specific liver and peripheral cell receptor, and is essential for the normal catabolism of triglyceride-rich lipoprotein constituents. This gene maps to chromosome 19 in a cluster with the related apolipoprotein C1 and C2 genes. Mu...
