Primary Abstract & Repository Description

Source: bioRxiv (Neuroscience)
The existence and mechanistic bases of sex effects on human subcortical anatomy remain unclear. To address these gaps, we combined structural neuroimaging of sex differences in subcortical volume and shape across three independent cohorts (combined N=2,886) with neuroimaging maps of sex chromosomal and endocrine effects from three clinical cohorts (combined N=313, including sex chromosome aneuploidies and isolated gonadotropin-releasing hormone deficiency). We found that human subcortical anatomy shows focal sex differences of moderate-strong effect size during young adulthood (male-biased surface area overlying lateral/ventral putamen and pallidum, nucleus accumbens, ventral posterior complex of thalamus, anterior hippocampus, and medial and central nuclei of amygdala; female-biased surface area overlying caudate head, posterior/dorsal pallidum, mediodorsal, lateral dorsal, lateral geniculate, and medial geniculate nuclei of thalamus, posterior hippocampus, and basolateral and lateral nuclei of amygdala). These regional sex differences show strong within-sample reproducibility at modest sample sizes (N[≥]200) and are replicable in a fully independent mid-adulthood cohort. Strikingly, we find that the topography of adult subcortical sex differences is already established by pre/early puberty (age 9 years), and the same holds for cortical sex differences. Comparison with neuroimaging data from rare disorder cohorts reveals that subcortical sex differences show a statistical...

Editorial Critique, Methodological Value & Reusability Guide

Structured via CNS Domain Knowledge Engine

1. Scientific Context & Premise

Primary Premise (bioRxiv (Neuroscience)): "The existence and mechanistic bases of sex effects on human subcortical anatomy remain unclear." Within Amyotrophic Lateral Sclerosis (ALS) & Motor Neuron Disease, this work examines bioRxiv Preprint, neuroscience, ALS in relation to plasma/CSF Neurofilament Light Chain (NfL), C9orf72/SOD1/FUS genetics, cryptic exon biomarkers, and ALSFRS-R slopes. Authored by Lee, H. M., Liu, S., Lalonde, F..

2. Study Architecture & Findings

Study Architecture: Experimental / Clinical CNS Design (primary study cohort) utilizing Preprint Article, Open Science [DOI: 10.64898/2026.10.05.756728]. Reported quantitative parameters in the abstract include: N=2,886; N=313; 9 years. Key reported conclusion: "Comparison with neuroimaging data from rare disorder cohorts reveals that subcortical sex differences show a statistical..."

3. Secondary Analysis & Reusability

Secondary Analysis Applications: (1) Cross-referencing Preprint Article, Open Science variables (bioRxiv Preprint, neuroscience, ALS) against open repositories in Amyotrophic Lateral Sclerosis (ALS) & Motor Neuron Disease; (2) Benchmarking feature distributions and effect sizes reported in "Sex Differences in Human Subcortical Anatomy are Robust and Replicable in Adulth"; (3) Informing cohort stratification and endpoint selection.

4. Methodological Caveats & Governance

Preprint Status: This record is an unrefereed preprint; findings should be verified against subsequent peer-reviewed publication. When integrating these Preprint Article, Open Science findings, account for cohort-specific inclusion criteria (primary study cohort), potential site/instrument batch effects, and repository license terms (cc_by_nc_nd).
