Patch-seq

Patch-Clamp Electrophysiology Combined with Single-Cell RNA-seq

Patch-seq(ホールセル・パッチクランプ電気生理+単一細胞RNA解析+3D形態再構築)

Multimodal single-cell technique simultaneously capturing intrinsic electrophysiological properties, 3D axonal/dendritic morphology, and full-length transcriptome from the same neuron.
Target CNS Conditions:Broad CNS / NeuroimagingAlzheimer's & DementiaParkinson's & MovementALS & Motor Neuron

1. Biological & Technical Mechanism

Patch-seq integrates whole-cell patch-clamp recording with intracellular dye biocytin filling and aspiration of the cell's cytoplasmic/nuclear contents for single-cell RNA sequencing. This bridges the historic divide between physiological/anatomical neuron classification and molecular transcriptomic cell-type taxonomies (MET-types: Morpho-Electro-Transcriptomic types).

日本語での専門解説 (Japanese Reference):

2. Open Science & Clinical Trial Significance

Massive open Patch-seq datasets from mouse and human neurosurgical cortical tissues are hosted on the DANDI Archive in NWB format and cross-linked with the Allen Cell Types Database.

日本語での臨床・データ活用文脈 (Japanese Context):

Related Neuroscience Terms & Standards

NWB — Neurodata Without Borders (NWB v2.x Neurophysiology Standard)

Unified data standard for cellular neurophysiology, packaging intracellular/extracellular recordings, optical physiology, and behavior into HDF5/Zarr files.

scRNA-seq — Single-Cell & Single-Nucleus RNA Sequencing (scRNA-seq / snRNA-seq)

High-throughput transcriptomic profiling at individual cell or nucleus resolution, enabling identification of disease-associated microglia (DAM), astrocytes, and vulnerable neuronal subtypes.

EEG — Electroencephalography & Intracranial EEG (EEG / iEEG / LFP)

High-temporal-resolution recording of postsynaptic cortical electrical potentials and neural oscillations (delta, theta, alpha, beta, gamma bands).
