SNCA

Synuclein Alpha (α-Synuclein / SNCA Gene)

Presynaptic neuronal protein encoded by SNCA whose misfolding and fibrillar aggregation form Lewy bodies in Parkinson's disease and Lewy body dementia.
Target CNS Conditions:Parkinson's & MovementBroad CNS / Neuroimaging

1. Biological & Technical Mechanism

Alpha-synuclein (encoded by SNCA on chromosome 4q22.1) is an abundant 140-amino-acid protein localized primarily to presynaptic terminals, where it regulates synaptic vesicle trafficking and neurotransmitter release. Point mutations (e.g., A53T, A30P, E46K) and gene multiplications (duplications/triplications) of SNCA cause autosomal-dominant familial Parkinson's disease. In sporadic Parkinson's disease and Multiple System Atrophy (MSA), soluble monomeric α-synuclein converts into beta-sheet-rich oligomers and fibrils.

(Japanese Reference):

2. Open Science & Clinical Trial Significance

Recent breakthroughs in CSF Alpha-Synuclein Seed Amplification Assays (αSyn-SAA / RT-QuIC) allow biochemical detection of synucleinopathy in prodromal cohorts (such as PPMI and MJFF pipelines). SNCA expression and splicing isoforms are heavily studied across single-cell atlases and DANDI electrophysiology models.

(Japanese Context):

Hunger reconfigures a reward learning circuit into a memory competent mode

Internal states such as hunger dynamically reshape activity across circuits to support resource seeking. Neuromodulation provides a means of controlling such physiological properties of neurons, but how this flexibility regulates memory networks remains unclear. Here, we describe how hunger reconfig...
